The quality of the DNA is of major importance in our analysis. Our manipulations require high quality genomic DNA, and we ask for a total of 2ug DNA from each pool (F2 mutant and parental). Degraded DNA that we often see with lysis-extraction protocols has often yielded poor results using our technique. A recommended DNA isolation protocol is listed here. Ideally, we want the pooled genomic DNA from this preparation to be concentrated to at least 10ng/ul.
