Targeted Sequencing

Partnered with GTSeek LLC

WHAT IS Targeted-SEQ?

Targeted Seq, or targeted sequencing approaches, assay predetermined sites within a genome in a highly multiplexed manner. Targeted Seq examines a preselected subset of a genome, enabling high-throughput applications such as genotyping by sequencing. This approach allows researchers to focus on specific regions of interest, such as known single nucleotide polymorphisms (SNPs), disease-causing genes or genomic regions associated with a particular phenotype. Targeted sequencing can be performed using a variety of techniques, including PCR-based amplification, hybridization capture, and amplicon sequencing. It requires precise and prior knowledge of the genomic sequence around the variants or regions to be assayed.

Floragenex is pleased to partner with GTSeek LLC (https://gtseek.com/) to deliver targeted sequencing services with the Genotyping by Thousands technology and Nate's Plates (https://gtseek.com/product/natesplates96w10/)

APPLICATIONS

Targeted sequencing can be employed in a high-throughput manner (thousands of targeted loci across thousands of samples at one time) to consistently assay individuals for specific sequence variants previously identified through RAD-Seq, WGS, or other techniques. Common uses include:

  • Environmental ecology studies

  • Targeted exome sequencing projects

  • Conservation and species validation studies

  • Molecular breeding applications

EXAMPLE Targeted-SEQ PROJECT

Genetic Mapping, Phylogenetics, Molecular Breeding

Surveyed >20 livestock samples

Used two step PCR amplicon based sequencing, on a 200-400 bp region of interest

Sequenced on the MiSeq 2x150, nano

Attained 100x+ sequencing coverage, scalable for high multiplex variant detection or rare allele event detection
